Depends if there is a need to test L-N. If you are testing new work then a full test between live conductors and lives to earth should be carried out. If it's an EICR or a test on an existing circuit where you are not entirely sure of the layout then a LN-E test is the only practical way, in which case there is no reason to disconnect loads. Try disconnecting 20 downlights before carrying out a L-N test......I don't think so.

Depends. Many countries only require a Live/Earth insulation test rather than a Live/Live one. Generally it's the insulation resistance to Earth which we are trying to ascertain for the installation, although obviously in some places it's between all conductors.
 
If you need to test between line and neutral then yes connected loads need removing/isolating. If you are carrying out some kind of periodic inspection then generally a line to neutral test isn't really necessary.
 
Doing the L-N test with an approved calibrated tester with any load will do one of two things....

1. give you false readings, as the tester is measuring resistances etc through the equipment.

2. severely damage the connected equipment.
 
If there is a desire to test an existing circuit L-N always do a 'soft' 250v test initially. That way you wont fry the clients £2000 PC which you didn't realize was on that circuit.
